تاثیر ژنتیک، نوع ریزنمونه، تنظیم کننده‌‌ رشد گیاهی و بستر کشت بر باززایی درون شیشه‌‌ای رقم‌های تجاری ارکیده فالانوپسیس

ارکیده فالانوپسیس (Phalaenopsis)، محبوب­ترین جنس‌‌ تیره ثعلب‌‌ها می‌‌باشد که به دلیل داشتن گل‌‌های پروانه‌‌ای شکل و جذاب، بازار پسندی بالایی دارد. پژوهش حاضر با هدف بررسی اثر ژنتیک، ریزنمونه، تنظیم کننده‌‌های رشد و بستر کشت بر افزایش درون شیشه‌‌ای ارکیده فالانوپسیس انجام شد. تیمارها شامل چهار تیمار تنظیم کننده رشد بنزیل آدنین (2 و 4 میلی‌‌گرم بر لیتر BA) و ایندول بوتیریک اسید (5/0 و 1 میلی‌‌گرم در لیتر  IBA)، دو محیط کشت MS و Chen، دو ریزنمونه برگ و ساقه، و پنج رقم تجاری فالانوپسیس بود. عوامل مورد بررسی تاثیر معنی‌‌داری بر زمان تشکیل سرآغازه‌‌های برگ و ریشه، ایجاد پداژه‌‌نما، شمار برگ و ریشه داشتند. کمترین زمان لازم برای تشکیل نخستین سرآغازه‌‌های برگ و ریشه، مربوط به رقم‌‌های  Nottingham و Dubrovnik، تیمار 2 میلی‌‌گرم بر لیتر BA ترکیب با  IBAبا غلظت‌‌های 5/0 و 1 میلی گرم بر لیتر، و محیط کشت Chen بود. زمان تشکیل سرآغازه‌‌های برگ در ریزنمونه‌‌های برگی در مقایسه با ریزنمونه‌‌های ساقه، به‌طور معنی‌‌داری کمتر بود. ریزنمونه‌‌های برگی، پداژه‌‌نمای بیشتری نسبت به ریزنمونه‌‌های ساقه تولید کردند. در رقم Nottingham، تیمار BA+IBA با نسبت 2  به 1 میلی‌‌گرم بر لیتر و محیط کشت Chen بیشترین تاثیر را بر افزایش شمار پداژه‌‌نماها داشتند. افزون بر این، شمار برگ و ریشه در گیاهچه‌‌های تولید شده در شرایط درون شیشه‌‌ای، به‌طور معنی داری زیر تاثیر عوامل مختلف قرار گرفت به‌طوری که گیاهچه‌‌های رقم  Dubrovnik شمار برگ بیشتری و گیاهچه‌‌های رقم Nottingham شمار ریشه بیشتری در مقایسه با سایر رقم‌ها داشتند. بیشترین شمار برگ در تیمار BA با غلظت 2 میلی‌‌گرم در لیتر به همراه IBA با غلظت 5/0 میلی‌‌گرم در لیتر به‌دست آمد. در حالی که بیشترین شمار ریشه در تیمار BA+IBA با نسبت 2 به 1 میلی‌‌گرم در لیتر وجود داشت. در مقایسه محیط کشت‌ها نیز محیط کشت  Chenتاثیر بیشتری بر تولید برگ و ریشه داشت. این نتایج نشان داد که موفقیت باززایی رقم‌های مختلف ارکیده فالانوپسیس در شرایط درون شیشه‌‌ای، وابسته به ویژگی‌‌های ژنتیکی، نوع محیط کشت و تیمارهای تنظیم کننده رشد است. رقم Nottingham و ریزنمونه‌‌های برگی باززایی بهتری در شرایط درون شیشه‌‌ای داشت. افزایش غلظت تنظیم کننده رشد سیتوکنینی BA از 2 به 4 میلی‌‌گرم بر لیتر، منجر به افزایش زمان مورد نیاز برای تشکیل سرآغازه‌‌های برگ و ریشه، و طولانی تر شدن دوره کشت درون شیشه‌‌ای شد.
