مقایسه روش های ازدیاد درون شیشه ای و برون شیشه ای سه رقم بنفشه آفریقایی (Saintpaulia ionantha H. Wendi.)

بنفشه آفریقایی ( ionantha H. WendiSaintpaulia.) یکی از معروف‌ترین گیاهان زینتی با رنگ­ ها و اشکال متنوع گل است. در این پژوهش تکثیر درون و برون شیشه­ ای این گیاه با یکدیگر مقایسه شد. در تکثیر درون­ شیشه­ ای اثر پنج ریزنمونه (پهنک، دمبرگ، دمبرگ بدون اپیدرم، گل کامل و گل بدون گلبرگ) در سه رقم (‘Cool Blue’، ‘Rondita’ و ‘Knight Rider’) در محیط­ کشت MS حاوی mg/l 1/0 NAA و mg/l 1 BA بررسی شد. در تکثیر برون­ شیشه­ ای قلمه­ های برگ سه رقم بنفشه آفریقایی درون ظروف پلاستیکی دردار حاوی 70% پیت ­ماس و 30% پرلیت کشت شدند. درنهایت سرعت تکثیر و تعداد جوانه گیاه در دو روش مقایسه شد. بررسی نتایج به ­دست آمده نشان داد که در هر رقم نوع ریزنمونه مناسب برای تکثیر گیاه متفاوت است. در رقم ‘Cool Blue’ هیچ­کدام از پنج ریزنمونه تفاوت معنی­ داری با یکدیگر در میزان باززایی، تولید جوانه و تعداد برگ گیاهچه­ های حاصله نشان ندادند. در رقم ‘Rondita’ ریزنمونه ­های پهنک (66/91%)، دمبرگ (66/91%) و دمبرگ بدون اپیدرم (33/83%) و در رقم ‘Knight Rider’ ریزنمونه ­های برگ، گل­ کامل، گل بدون گلبرگ و دمبرگ (همگی 100%) بیشترین میزان باززایی را نشان دادند. در تکثیر قلمه برگی رقم ‘Cool Blue’ بیشترین (25/8 عدد) تعداد گیاهچه را تولید کرد. مقایسه طول دوره کشت در شرایط درون و برون­ شیشه نشان داد که در هر سه رقم موردبررسی، دوره زمانی از کشت تا گلدهی در شرایط درون­ شیشه­ ای (180 تا 230 روز) طولانی ­تر از برون­ شیشه ­ای (120 تا 140 روز) بود، در حالی­ که تعداد جوانه تولیدشده در کشت درون­ شیشه ­ای به­ طور معنی­ داری در مقایسه با تکثیر قلمه ­برگی بیشتر بود.
